MOTS-c is a 16-amino-acid peptide encoded by mitochondrial DNA, discovered in 2015 by Changhan Lee and Pinchas Cohen (University of Southern California) and published in Cell Metabolism — the first to show that a mitochondrial sequence can act as a systemic hormonal signal. It is not an approved drug. Available evidence is mostly preclinical (mice, cell cultures), where MOTS-c activates AMPK, stimulating mitochondrial biogenesis, lipid oxidation, and glucose uptake in skeletal muscle. In 2026 it began its first human clinical trial (phase 2a, prediabetes/overweight), results not yet published.
