Origin, mechanism, and research

Developed at Saint Louis University (the Thomas Burris and Patti labs) and first described in 2022, SLU-PP-332 is a small synthetic molecule — not a peptide — nicknamed the "exercise mimetic." Its regulatory status is exclusively preclinical: all available efficacy data comes from mouse studies; it has not been replicated in humans, and no clinical trials are registered. It works as a pan-selective agonist of the three estrogen-related receptors (ERRα, ERRβ, ERRγ), with greater affinity for ERRα, activating oxidative-phosphorylation genes, beta-oxidation enzymes, and mitochondrial biogenesis. In mice, it increased oxidative muscle fibers and improved aerobic endurance capacity without the animal exercising.
